It was completely obvious Ron was joking. He wasn't even doing his bad acting, "it's a bit for you guys who are paying attention" voice. It was a straight up joke. He even sarcastically said, "On his last day of broadcasting ever, Letterman is doing Opie's show."

Opie was misquoted. He was meant to announce himself being the guest host of The Late Show with Opie which will air in between Letterman's retirement and Colbert's start in September. Part of the reason the Stangel's were hired, is to coach Op in his monologue delivery. He clearly has the comedic timing and interview skills already so he will need no practice in these areas.
